Is it possible to set up replication from Single-Filegroup to a
Multi-Filegroup?

Yes. You will need to use a creation script parameter to create the objects
on the subscriber in the file groups you want. Or, you could precreate the
schema on the subscriber and then in each article you can use the name
conflicts setting to keep the existing table unchanged.
